1. A type I AFP was isolated and purified from cunner (Tautogolabrus adspersus) plasma. It shows a seasonal variation with the highest activity in winter. The TH activity is 0.82℃ at 10mg/mL, which is higher than some other fish type I AFPs, and it causes ice crystals to form a hexagonal bipyramidal shape.
2. This protein is a classical type I AFP. It is rich in alanine, has three 11-amino acid repeat regions , has a high α-helical content (99.8%) with reversible thermal denaturation, and its secondary structure and ice-binding properties are similar to other type I AFPs. The molecular weight is approximately 4095.16Da (with post-translational modifications).
3. Partial sequencing of the plasma AFP and analysis of a combined liver and skin cDNA library indicated that the protein lacks a signal sequence. These findings support the hypothesis that type I AFPs have a multiphyletic origin and exemplify convergent evolution in teleost fish.
